Content Determination Method
Chromatographic Conditions: Silica gel G thin layer plate; Cyclohexane-chloroform-ethyl acetate (20:5:8) was used as the developing agent and developed upward. Spread distance 12 ~ 18cm; 5% sulfuric acid ethanol solution was heated at 110℃ for 5min to develop color.
Preparation of Reference Solution: Accurately weigh the appropriate amount of Ursolic Acid reference product, add the mixed solution of anhydrous ethanol: ethanol (3:2) to make a solution containing 1mg per milliliter as the reference solution.
Preparation of Sample Solution: Finely weigh and pulverize 20g of gardenia (fried gardenia according to yield conversion and weighing), put it in Soxhlet extractor, add 300ml of ether to reflux extraction until colorless, recover solvent to dry, and soak the residue in petroleum ether twice, 15ml each time, about 2min, pour petroleum ether, and use anhydroethanol-ether (2: 3) slightly heated and dissolved in a 5ml bottle as a sample solution.
Determination: Accurately absorb the sample solution and the control solution 2μl, and point on the same thin layer plate. Ursolic acid can inhibit the production of superoxide induced by formylmethionyl-leucyl-phenylalanine (fMLP) and arachidonic acid respectively. When ursolic acid and fMLP were added to the cell culture medium, ursolic acid inhibited the tyrosine phosphorylation of 45ku protein induced by fMLP in a dose-dependent manner. Ursolic acid also has trypanocidal activity. After incubation for 2 hours at a concentration of 2 g·L, the activity of Trypanosoma cruzi's flagellates was completely inhibited.
